What is everyone using as floor mats in histogy.  The mats we have are 
very thick and with Holes,  we are finding it difficult to move the chairs 
on them to get close to the microtome for cutting.  We were told that 
carpet of any kind cannot be use.  If we remove the mats, the floors are 
very slippery.  Any idea will be greatly appreciated.

We had this problem several years ago.  We  were using the sakura tapes 
with the coverslipper. 
We did the following: 

Last three alcohols were changes frequently. 
Slides should be not dry when loading on coverslipper.
If you could load two racks at a time, only load one.  By this way the 
slides in the second rack will not dry out. 
Finally, change the tapes from sakura to Mercedes Medical tapes.

We also have recently started to see this artifact more than ever before, 
and nothing in our process has changed.  We have tried everything to 
correct to no avail.  Wonder if it is possible to be a change in some type 
of supply, either xylene or coverslipping film.  Something has changed but 
am at a loss as to what.

You will also see the cornflaking if your tissue is lifting off of the 
slide at all.  We used to get this more often on hard, decal specimens 
than on other specimens.  We used the film to coverslip.  If you remove 
the film from the problem slides and recoverslip conventionally with extra 
mountant and glass coverslips, I'm sure you will not see the artifact.

I am looking for help with cornflaking (tiny, brown dry spots under 
coverslip)artifact.  We have been using fresh xylene on our stainer and 
coverslipper, cleaned and wiped all containers dry before filling, tried 
different lots of coverslipping film and had service on our coverslipper 
to make sure it was functioning properly, including the xylene drip.  We 
continue to have this artifact and it is driving us crazy.  It is sporadic 
with no pattern of tissue type or placement on the slide.  Sometimes it 
lands on tissue other times not.  Most of the time when we remove the 
coverslip and re-coverslip it goes away (I am assuming because the acetone 
removes any minute amounts of water that may be present).  We just cannot 
figure out where the water is coming from.  Has anyone seen this artifact 
while using the drying step on the prisma stainer?  We just recently 
started using the drying on some slides and I am thinking maybe it is 
causing humidity???  I cannot say for a fact that our cornflaking 
started at the same time, but it is suspicious. HELP!

We are all putting out what worked for us.  You should  try some of these 
suggestions and see what works for you.  The suggestion for improving poor 
dehydration was suggested by technical support  from Sakura.  Cornflaking 
was irritating so many of our pathologist, we had to get technical support 
from Sakura to see how this could be resolved.  In OUR hospital changing 
the tape worked!!!  Good Luck.!!!

 We had this problem several years ago.  We  were using the sakura tapes
 with the coverslipper.
 We did the following:
 Last three alcohols were changes frequently.
 Slides should be not dry when loading on coverslipper.
 If you could load two racks at a time, only load one.  By this way the
 slides in the second rack will not dry out.
 Finally, change the tapes from sakura to Mercedes Medical tapes.

 You will also see the cornflaking if your tissue is lifting off of the
 slide at all.  We used to get this more often on hard, decal specimens

[Histonet] knife used for cleaning paraffin off the blocks

I wonder if someone could help me with this!

I am looking for a stainless steel knife that we use to scrape off the 
paraffin from the embedded blocks.  I could not find the 6 inches size 
knife in any catalog.

[Histonet] problems with staining IHC

Hi All,

Our pathologists have made a complain that the staining of slides have 
been a problem.  Same antibodies which stain one day does not work the 
following day.  This has been going on for a few months.  When we repeat 
stain they either work or not work and we have been sending them to 
reference labs. 

Our tech support person was here and told us that the slides ( fisher plus 
slides) have been a problem with some of her customers.  Meanwhile our 
surrounding labs which are using the same slides have no issues.  She 
showed us how the reagents were not spreading properly on the slides. 

Is anyone confronted this issue?  If so,  how was it solved?

Hi All,

We are using Ventana XT for our immunostaining.  We tried using Plus 
slides made by many vendors.  The current vendor's pricing is way too 
high!  With other vendors slides  we have been having problems.  Please 
let me know where to get some plus slides which are reasonable.  I know 
the recommended slides come from erie but I am not quite sure about 
vendors who carry it and how much they charge.

Ventana Users,

Need some advise regarding the RUO and ASR antibodies.

Is anyone out there in a hospital setting using RUO and ASR reagents with 
the Ventana XT?  If so,  is it necessary to add a disclaimer on the 
reports?  Are they chargable?  Are you using IVD antibodies from other 
vendors? 

The antibodies under consideration are p63 and mammoglobin.

[Histonet] regarding using neg controls for H.pylori

Good Morning,

I am sure this question has popped up before, but,  when you are running 
H.pylori by immunohistochemistry do you have to run a patient pos and 
patient neg for each patient.  If you have 10 patients do the run consist 
of 20 slides each patient having a pos and neg. What does the CAP suggest 
us to do.?  We have a debate going regarding this issue and I would like 
clarification.  I understand when we do 2 slides each  there is only one 
billable item and the neg is using reagents but cannot be reimbursed.

[Histonet] XXXL Latex Gloves

Hi,

Does anyone know a company who makes XXXL latex gloves?  We do have a 
morgue  attendant who claims the XXL gloves that we purchased(after a very 
long search) is too tight.  Any help will be greatly appreciated.

What are the special stain chemicals that need to be shipped out as 
Hazardous Waste and not go down the drain into the sewer system. We know 
silver,chromic,gold,formalin and alcohols.
What about 30 ml of Carbol Fuschsin solution or  working iron stain?
Any clarification on this will be helpful.

We have a morgue attendent who does not fit into the XXL gloves.  Does 
anyone know a vendor who can provide XXXL GLOVES latex (powder free) 
prefered, but will take any other brand too.

You can view the MSDS for CBG Biotech's Formula 83 xylene substitute at

It's described as a naphthenic hydrocarbon blend.

I'd note the low flash point (7 C, 45 F), considerably lower than
xylene's. Other aliphatic or naphthenic (cycloalkane) mixtures offered
as clearing agents have considerably higher flash points.

Remember that if you want to recover it by distillation, you must make
sure that your still has a distillation routine for it, and you cannot
change clearing agents without changing the distillation routine, nor
can you mix the solvents and recover them by distillation.

If your lab manager shares cost information with you (in my experience
most do not) then cost will certainly be a consideration, if it's
indeed cheaper than xylene.

According to the MSDS, disposal is done like any other aliphatic or
naphthenic clearing agent.
