[Histonet] Air Bubbles
Has anyone encountered the following problem. Tissue cut and is processed well. There is no distortion at a cellular level. However anywhere from 30 to 2 days later there are air bubbles under the coverslip. I know for certain there aren't even the slightest bit of air bubbles when coverslipped. I have a feeling it could still be a processing issue as I ran a few tests like coverslipping blank slides and tissue sections from a year ago when there was no issues. There might be air in the mounting media that gives rise to bubbles when drying starts to occur. One method to try is to use an ultrasonic bath to remove the air from the bottle containing the mounting media before using. We used to do this with aqueous mounting media. [Histonet] Air Bubbles
Hello everyone, Recently we have been having an issue with a majority of our slides having air bubbles under the coverslips. When they are originally coverslipped there are no bubbles and any present are pushed out and the sides are all wiped dry. We have not changed any reagent or processes in over two years. Hi, We always have more than one medium-bottle prepared. At least three are filled in advance. So entrapped air can go away until usage and the oldest is the one for exchange. I do not use an automatic cover-slipper so don't know if this will help. However the delete button is nearby. When we wanted to remove trapped air from any solution we gave the container a short period in an ultrasonic bath - 10 seconds or so (depending on viscosity of the solution. This worked well for some of the media we used before embedding in electron microscopy. We have flushed the hose from the mounting medium through the needle with xylenes which sometimes works to clear the bubbles and sometimes does not. Has anyone else had trouble with this and possibly found a solution? I would really appreciate your advice!
