Re: [Histonet] I have a couple of quick questions
Yes. to remove mounting media you need to leave them in the Xylene after the coverslip is removed. If the mounting media is not totally removed your stain, whatever one you are doing, will not give you the results you need. Beatrice Sullivan, HT(A.S.C.P.) HTL , AAS, CLSP(N.C.A.) Do I need to use xylene to get rid of the semi hardened permount gunk? Also, If I am heating paraffin sectioned slides overnight at 37C to adhere them what happens if I over cook them by leaving them at 37C for more than one day? Say 3-4 days at 37C It probably wont hurt them as they haven't been epitope retrieved yet. Yes, xylene is the way to go. No, 37ºC will not hurt the sections no matter for how long they are at that temp. Yes, soak slides in xylene to remove coverslip. A few days in 37 degree oven won't hurt Roxanne Sent from my Verizon Wireless Phone - Reply message - From: Lewis, Patrick patrick.le...@seattlechildrens.org Date: Thu, Oct 14, 2010 7:11 pm Subject: [Histonet] I have a couple of quick questions To: Histonet@lists.utsouthwestern.edu If I want to remove a cover slip that is coversliped with permount.
