No, Designer doesn't support the #region directive, just like it is
not supported in .aspx files in studio.
 
I believe these sorts of directives can be used in pre-compiled code,
whereas aspx files are not compiled, but are interpreted by IIS as is.

I think you can put anchors or some sort of marker that will allow you
to move to different points within your code. 

This might help you navigate it.
