It sounds like you are having a problem with the authentication method,
so I think the best way of finding out what is going on is by installing
a packet sniffer (like Wireshark) and seeing what is happening at the
packet level. Then, if the packets show the correct user info being
provided to the domain controller for auth, and the DC fails to auth,
check the security eventlogs of the DC for the reason why.  That should
give you a clear idea of what is going on.

What  you expect wrong with app pool ? As I said if you set
authentication type to basic and provide a user name and password in

The page is just a test page and only have a calendar view other than the data 
view.. Also I was testing with SharePoint point web service and it works fine 
even I set  authentication type none, is it for SharePoint web service designer 
pass the authentication implicitly?

outside the sharepoint box? Does it connect to any datasource on another box,
