Well, the style file itself declares:

"This is a numerical citation style, and as such is standard LaTeX."

So I suppose it is not possible.

Thank you for this help. I can see the bst file plos2009 now. However, if I
choose "Standard (numerical)", references are numbered and not by
alphabetic order as needed by the journal. How to go through this ?

Save the bst file in a directory where LaTeX can find it 
(.../texmf/bibtex/bst/plos/).

In the BibTeX inset dialog in LyX, hit "Rescan".

Now you should be able to select it from the "Styles" combo in that dialog.

Note that the file is not compatible with natbib or jurabib, so you need to 
select "Standard (numerical)"
