Sure, you can have separate config file.

You can specify as many config files as you want in your nagios.cfg.
cfg_file=/some/file

Alternatively you can specify directories in which all your *.cfg files will
be loaded.
cfg_dir=/usr/local/nagios/etc/server

yes, just source the extra files in your nagios.cfg like so

cfg_file=/path/to/some.cfg

Yes, certainly. Create a new file, move your desired hostgroup
definitions to it, include it as a cfg_file in nagios.cfg and restart
nagios.

I have used the cfg_dir. My Nagios config looks like:

cfg_dir=/usr/local/nagios/etc/hostgroups

In the directory, I have multiple hostgroup files.
