I _think_ you should be OK. I just did a similar thing with a SharePoint List, creating a template from an existing list, using the template to create a new list, then deleting the template. The list I created from the template still works OK. This was with SharePoint 2007, but I would expect 2010 would be the same (not taking any responsibility if it doesn't though! J )
