There may be two reasons:
1. Somehow, the queue structure (or access rights) got damaged. 
Use a tool which shows that, like "make setup check" or
"queue-fix" from www.qmail.org.
2. Besides being out of free space, you may be out of inodes. 
Check not only "df", but also "df -i".
