Correct. I've done so many times.

normal logs. I don't believe there is anything different in the logs
regarding smtproutes. Someone please correct me if I'm wrong on that.

The only way I can think of to know if its working is just to run a test.

Yes just

 qmailctl restart

AFAIK its just normal traffic that will display in the relevant log  
file.
