Try checking if the  /tmp/libgksu-*/.Xauthority file is created with the
right ownership of the target user?

I also get

Xlib: connection to :0.0 refused by server
Xlib: No protocol specified

on running 
$ gksu /usr/bin/update-manager

when the user is not in admin group but
user ALL=/usr/bin/update-manager 
is in the sudoers file

This is the same for any app I choose to put in the sudoers file.
