There's a TeX derivative called XeTeX (http://scripts.sil.org/xetex) that (on Linux and Windows) uses a modified version of ICU over fontconfig and freetype to access and "render" system-installed OpenType fonts. I put render in quotes since XeTeX doesn't really draw the glyphs, but it does need to determine font metrics etc. XeTeX supports both flavors of OpenType equally well, and it has *full* support for OpenType layout features: you can set any feature tag directly if you like and there's also an abstraction layer.
