The Cryodrive is already gone.

Dear all,
is anyone still using glass capillaries for crstal mounting at room
temperature ???
We have a couple of them in size from 0.2 - 2.0 mm diameter and 80 mm length.
I would be happy to give them away.
Best,
Ulrike
Hi Jon,
here in Germany we got the Honey Bee installed by Zinsser Analytic.
May be they are still doing the service .

You have to scroll down the page a bit to find Zinsser.
Good luck !

Hi Ursula,
you could try Perfluoropolyether PFO-X175/08 (Hampton Research HR2-814). You
just plunge your crystals in the liquid or wipe the crystal in the loop through
the liquid so that all excess water is removed.
Good luck !
Ulrike
Dear crystallographers,
I am trying to crystallize a soluble protein which tends to form aggregates.
The crystallization condition is 20% PEG 3350 + 0,2 M Na-Formate. During the
crstallization process a thick skin is formed on top of the sitting-drops. As
well the crystals are buried in
Hi everybody,
I want to buy a smaller incubator for protein crystallization (30-50 l). Do you
have any recommendations ? Do you prefer specialized incubator models with low
vibration or are you just using standard incubators ?
Best wishes,
Ulrike
Dear Careina,
altough your crystals does't take up the Izit dye it sounds promising. The
uptake of Izit depends on the solvent channels of the protein molecule -
sometimes the dye just can't enter the molecule.
Concerning the Calciumchloride - if the concentration is not too high and
without
Dear Mike,
we got a dry shipper from Taylor Wharton (
http://www.taylorwharton.com/default.aspx?pageid=72).
In Europe dry shippers and accessories are sold by Molecular Dimensions
(http://www.moleculardimensions.com/shopdisplayproducts.asp?Search=Yessppp=6).
Cheers,
Ulrike
Dear Judith,
in Acta Cryst D61 (2005), 1173-1180 different handling methods for dehydration
are described besides other post-crystallization treatments to improve
diffraction quality.
You can also try Dehydration Salts. A screen with ready made salt solutions is
available from Jena Bioscience.
Dear Shankar,
I once got crystals with Ethylene glycol as precipitant but at low
concentration (8-14%) and there always had been spots right from the beginning.
It sometimes happens that you get nice looking crystals but they do not
diffract. If you have hits from other conditions may be you
Thank you all for sharing your experience with Perfluoropolyether and freezing
membrane proteins !
Now I myself heard about one membrane protein where Perfluoropolyether was the
solution for the freezing problem. And it seems it worked in my case as well
:-)
Ulrike
Dear crystallographers,
does anyone have experience using Perfluoropolyether (Hampton Research) as
cryoprotectant for membrane proteins ?
Cheers,
Ulrike
Dear Eva,
are you using siliconized cover slips or normal ones ? Protein drops on not
siliconized glass tend to spread especially if they contain alcohol or
detergent. But this shouldn't be a problem with sitting drops usually.
Good luck !
Ulrike
Dear Ed,
try ca. 30 % PEG 400
Another possibility could be Perfluoropolyether Cryo Oil from Hampton Research.
This liquid doesn't mix with water. You put the crystal in few microliter and
remove the excess water droplets from your screen solution (otherwise you will
get ice rings).
Cheers,
Dear Nishant,
are you sure your crystals are protein ? The condition contains 1 M of
different salts and PEG as well - I wouldn't exclude that you are having salt
crystals. You can easy check this with Izit dye or simply by trying to destroy
the crystal with a microtool. If it is salt you can
Dear crystallographers,
does anyone know if dialysis buttons for crystallization are available
somewhere in Europe or only at Hampton Research ?
Thanks,
Ulrike
Hi everybody,
has anyone experience with Butandiol and its different isoforms as
cryoprotectant ?
Thanks,
Ulrike
Dear Paul,
AMP-PNP is available from Sigma-Aldrich. Product numbers are A 2647 and
01910.
Best wishes,
