This happened to us recently running on J2EE settings but started working
once JVM property coldfusion.disablejsafe=true was added.

Just applied hotfix 16 to CF10, now the application service won't start.

As a start, I would check your CF logs to see if there is any additional
information that may shed light on the issue.
Event logs are usually pretty generic when it comes to CF.
You could also roll back the hot fix to get things working again.
