Yes you can but it seems to be platform specific:
interface Serial4/1/2
no ip address
no ip directed-broadcast
no ip route-cache
no ip mroute-cache
loopback
C7500MIX#sh int ser 4/1/2
Serial4/1/2 is up, line protocol is up (looped)
Hardware is cyBus Serial

My finding is different with you. What IOS version do you use ?
This behavior may be version or router type dependent .

Guess I hit the exception!!
I tested in on a MC3810 running 12.1(9)!
Under the site you gave me it says that the command "loopback" is
modified for that router!!!
I'm glad we solved it now. ;-)

Use "loopback" command at the serial interface mode.
For ethernet I/F, use "no keepalive" command.

this will cut off the electrical signalisation
