After a recent upgrade of my whole system, I'm encountering the same problem here. It seems that the module snd-mixer-oss is no longer loaded by default. Loading the module by hand does fix the issue. I am not sure that is a bug in tkmixer though. It's probably supposed to work only for OSS.
