If you have only the GPO Finale edition, the file is called just
"gpoinstrument.txt", not "gpofullinstrument.txt".

Actually, if you require GPO playback for a score with a large number
of instruments (e.g., full orchestra), it's much, much faster to use
the Setup Wizard and edit as needed later. The reason is because the
Setup Wizard automatically configures your score for GPO playback,

Ah, you must be on Mac, then you need to use AU, not VST.

Perhaps a good test (or at least it may help in general
troubleshooting) is to ignore it in the wizard and see if you can turn
it on from the menus in the file.

If you are talking about the FULL GPO, not the Finale GPO, the file
PersonalOrchestraVST.dll must be copied into the Finale VST directory.
