Hi Cecilia,
Glad to know post fixation will reduce section falling off from slide. We are
bone research lab and always have problem of paraffin bone section falling off
after antigen retrieval. There are many ways we have tried, but not improve too
much. I know bone section will be easy to fall
Hi,
I couldn't do on paraffin tissue after trying a few times. But works fine on
MMA plastic section.
If any one has a practical protocol, please share. Thanks in advance.

Hi,
We are using Kawamoto's Section-Lab Co. Ltd. products now. It is better than
cryojane. Check Kawamoto's paper online to get protocol. Good luck.

Hi,
You may use formic acid instead of EDTA, it will be much fast and give you
sharp image of Safranin O. You can also do IHC.

Hi,
I make staining solution for TRAP staining. Used once Sigma kit, but
unsatisfactory. You may Google for TRAP protocol online. It must be there and
easier than the kit.

Hi histoneters,
Does any one out there to process tissue in a tissue process machine after lead
or barium agents introduced to your tissue? It will do any harm to the machine?
Get clog or contaminate other tissue? Thanks for your help.

Hi Susan,
I was so glad that I saw your post regarding tissue falling off. We have had
this problem for a while, especially for human bones tissue. This prevents us
to do any heat induced antigen retrieval. If it possible to email me how do you
mix soapy water with ammonia? How many ml of water
I try to avoid CaCl drying step in making MMA infiltration and embedding
solutions. Please help me MMA experts. Many lab use benzoyl peroxide which has
30% water in it. And go through CaCl afterwards.
Another question, why some lab use Butanol and toluene before coverslip? Why
use Cellosolve to
Hi Gayle,
Thank you very much. Please send me the paper of perkadox from Stain
Technology. We try to determine benzoyl peroxide or perkadox to do mouse
undecalcified bone in MMA.
Any previous people's study regarding this topic will be greatly appreciated.

I am new to MMA plastic bone technique. Some one gave me his protocol, in which
has NaOH and d-water to wash MMA mixture before drying it in CaCl2. But others
told me I don't need to do the destabilization step. Could any expert in this
area to tell me if this step is necessary? And why have to
