You did not specify which enzyme you were using, but I am assuming proteinase K. If an hour's retrieval is too much, and no retrieval is too little, I would suggest running some (alcohol fixed) controls at 15, 30, and 45 minutes in the 95C, or 1,2,3, and 4 minutes in the enzyme. Evaluate the
