Use a formic acid based decal solution and MAKE SURE the specimen does not
fix (formalin fixation) longer than 32hrs (false negatives result). Use 70%

Hi Kendra-
Decal Stat from Statlab is HCl based, therefore is not appropriate for
subsequent IHC, ISH, or other molecular studies. There is lots of
peer-reviewed published data, if you need to supply rationale to your
pathologists. HCl is known to significantly reduce molecular assay
Hello all,
Recently, the Kappa/Lambda ISH has quit working on our decal specimens.
When we first validated ISH, we tested decal specimens that stained
appropriately. We use the DECAL Stat from StatLab. I did NOT do the initial
ISH validation back in 2019, but as far as I know, we have not changed

Good morning!
Just recently we started having an issue with a heavy blue precipitate on
our bone marrow cores. We have changed nothing in our protocol and even
did parallel testing with a reference lab. Both had the precipitate, but
to a lesser intensity.
Has anyone else been seeing this?
I am looking for any info/webinar/educational material for in situ
hybridization (ISH). We would like to bring this into our lab on the Ventana
BenchMark Ultra.
Thanks,
Heidi

That may be what you have to do, as the first day of ISH needs to be RNase
free!
When I do them, it is 10 slides at a time and we have dishes and a lab
bench set aside for RNase free procedures only. Obviously this is not
feasible in all workplaces!
I would just think about it logically. Use
Hi Folks!
I was hoping to get a little input about preparing samples for ISH.
We currently RNAse-away our tools at the time of collection, toss the tissues
into formalin, process normally and then cut them using RNAse away cleaned

We are having sporadic issues with kappa and lambda ISH on our bone marrows.
What effect does EDTA have on ISH? The same as decal?

Would anyone care to share their ISH protocols? Especially ISH double
staining with DAB and Refine Red. I use the Leica Bond Max. I am not
having luck optimizing this protocol.
Happy Holidays!!

Hi Everyone,
We have some cells that we have processed and embedded in histogel (thanks to
everyone for the protocols that were sent to me a couple of weeks ago).
We are planning to do IHC and ISH on these pellets. Will the histogel
interfere with the ISH procedure at all?

Dear Histonetters,
I have a problem with my ISH and maybe one of you can help me.
I used probes I *know* they are working. I put the mice embryos (E 10.5)
in BM Purple for staining. Unfortunately they turned black in a short time.
They were not stained in the inside. It looks like that the

Hi Histonetters!
We have recently changed to Immunocal to decal bone marrow biopsies. Immuno
stains performed on the decaled bone marrows look good. All In-situ
hybridization negative controls, and Kappa and Lambda stains on decaled bone
marrows have a blue artifact at the edges. We use the

In our lab the unstained IHC-slides are thrown away. Now one slide of each

I want to store it unstained but deparaffinized and coverslipped. So that it
