After grossing in, we soak the nail in Nair (dab of HOT water mixed to
runny paste) for a couple of hours to soften the nail.
We face into the block, soak the face of the block in fresh Nair solution
again for at least 30 minutes.
Bake sections in the oven for 1 hour at least, sometimes hand stain
Does anyone have any special tricks to keeping nail sections on the slides? I
am doing a PAS stain and the tissue keeps falling off.
Using + Slides, cut on 1-2 microns, baking 1/2 hr. Air drying, and this nail
still wants to fall off.
Any suggestions. Please help,

Our lab receives toenails, and we routinely do an HE stain and a PAS stain for
fungus. We receive the nails fresh, and when they are grossed they are placed
directly into a purchased nail softening solution that is mainly potassium
hydroxide before being placed on the tissue processor. My
or else they will get washed off slides.

Histology
This helps to keep sections of bone from floating. I mount them on slides
coated with water resistant wood glue, then immediately microwave dry the
slides. 40% power for 3 minutes. No doubt there will be variations, depending
on the oven.

I use a 5% solution of Titebond 2 wood glue to coat slides. You should be able
to find it at your local hardware store. Works well for me on bone and bloody
tissue, Occasionally I need to microwave dry slides of very dense bone (skull,
inner ear).

An old old trick is taking a plain slide and coating it with dilute Elmer's
glue and letting it dry. Be quick
The least complicated way to cut nails is to face the block, get a small
shallow dish and put in a few drops of Dawn dish soap and add a little warm
water. Just soak for about 10 minutes. Cut and place on a + slide and dry
slide well before staining.
We are having a very difficault time with the naisl falling off the slides when

increasing the baking time during drying to no avail.
