Thank you for the setUseAscender(true). This works well now!

ps:Because when I print the itext pdf then measure the middle of the cell with my ruler, the text "middle" was not in the middle. And with word or internet explorer the text is exactly in the middle.

You may be missing a PdfPCell.setUseAscender(true). We can also argue why would you consider Word "more correct" than iText.
