At least on my servers, MIMEDefang uses the scanner daemon (clamd) via the socket configured in $ClamdSock. (/var/spool/MIMEDefang/clamd.sock by default.) Actually, perusing the Perl source in /usr/bin/mimedefang.pl I see that it seems to support both ways. Ie. if the ClamAV daemon is not available it will call clamscan directly. However I would recommend against it from a performance point of view.
