I've got the same problem and a suspicion as to the cause. I installed
the multiload indicator applet on my machine and I've only noticed the
problem occurring since I did that. Might be unrelated, but seems like
it could be part of the problem. Does that sound relevant to your
situation?

After purging and installing alsa-base and pulseaudio, my sound is back
and volume can be adjusted using the slider. I will pay attention to
what I'm doing if it happens again. Marking as invalid for now.
