I tried enabling it as well and it seems to work:
$ echo CHECK_RCPT_SPF=true > /etc/exim4/conf.d/main/00_local_macro
$ update-exim4.conf --verbose --check
using non-split configuration scheme from /etc/exim4/exim4.conf.template
All ok, so make it effective.
