Nessus is very good, but for full control over your packets I recommend hping (http://www.kyuzz.org/antirez/software.html). This will allow you to create the most weird and wonderful packets that you could possibly need at the speed you need. I also use hping to do manual port scanning for those rare pen tests where you just can't get enough information from a host using, I hate to say this, nmap.
