No, DNS don't do that.
If this is for a web server, you'll need to tell people to use
the URL <http://happy.thanksgiving.com:yyyy/>. Which, of course,
will cause their heads to explode in fits of confusion.
What problem are you trying to solve? Do you not have access to
port 80 on the server? Or do you only have a single IP address,
and there is already another server on port 80?
If the latter, are you aware that these days most web clients support
the HTTP 1.1 Host: header, which allows you to setup multiple virtual
hosts on a single IP address?
