If the machine is already running ssh, then just install winscp client and attempt to send the big file via SCP protocol and see if the problem happens. As others have said if its network or nic related it should fault on any protocol including ftp, samba, scp etc

Interesting problem by the sounds of it though.

Just a suggestion. To determine whether it's something to do with
Samba or not, how about sticking a FTP daemon on the server and
uploading the big file through FTP? Would be interesting to see if it
works fine, then at least you'll know where the problem is.
