The differences in basic administration between 2000 and 2005 are minor,
though the interface is somewhat different and the Enterprise Manager and
Query Analyzer have been replaced with SQL Server Management Studio some
other tools are different too.  They aren't hard to use, just be prepared
for some minor adjustments.

Aside from the technical differences, I would definitely take into account
that if your organization if cheap and slow to upgrade, it can pay off to
make an argument for the most current version of everything at the start of
a project because you are likely to have problems getting it upgraded
later.  If this is for a dev environment, just make sure they are also
willing to make that same investment for the production environment.
