I would prefere
cache_dir aufs /var/spool/squid 10000 16 256 #Might need ./configure make in
case your squid

not built with aufs storage module
Should be faster under load. Then, you might be even able to increase
throughput by using more threads for I/O with option
--enable-async-io=nn
for configure, after checking the statistics with cachemgr.
