I am still somewhat new to TinyOS too.

In order to build the application, you have to go inside the directory of
Blink.
In a terminal in ubuntu, you would write: cd /opt/tinyos-2.1.0/apps/Blink/
(if you installed tinyos in the directory /opt)
Then you can use your make command ("make tmote install,1 bsl,/dev/ttyUSB0"
).

I am not sure if the tmote is still supported, but it gets replaced by
telosb at compile time, so I guess you should be fine.
