Yes. To confirm a mail, you can send absolutely anything at all to the confirmation address that TMDA generated for that email. TMDA only checks the address that the confirmation response is sent to - it doesn't look at any headers, or the body of the email.

Just the destination email address.
So if I send manually a mail to this address this should also be confirmed?
If you mean, send a manual email to the confirmation address, instead of pressing "reply" to the original confirmation request email, then yes.
