A have the same problem on Ubuntu 9.10 x86-64 Karmic, Rhytmbox plays .mp3, but there is no sound. In System->Preferences->Sound Applications: No application is currently plaing or recording audio. For example Totem plays .mp3 properly and it is visible in Applications.
