I think this information may be useful: I'm running Karmic's stock Wine with the ESounD driver just fine (i.e. PulseAudio's ESounD emulation rather than wine-pulse drivers). I had the same ALSA issue as others, I also tried Wine 1.2 + Wine-Pulse from PPA, but I'm back to 1.0.1 because 1.2 doesn't play ball with the window manager in Karmic.
