This involves about 60% manual testing and 40% automated. Manual testing is more about being thorough while automated is more on the technical side. Automated would include use of tools like QTP (scripting) and Winrunner. There are times when the Tester is required to write SQL statements (mid level not highly complex) to get certain data from the database (so some knowledge about databases is essential) in order to verify the test cases. Depending on the database Oracle / SQL server, the tester needs to be (again mid level) conversant with the tools that are used to connect to the databases and write the SQL queries in. So it is not highly technical but they need to know the tools used and scripting languages used to do the testing. That's about 80% of the job, the remaining 20% would be creating test plans, meeting with the business + business analysts + managers (communication skills), generating test reports in QC (HP has a tool called QC Quality Center).
