You can use WFS. Create an "intersects" (or "contains") spatial with filter. See <http://dev.openlayers.org/apidocs/files/OpenLayers/Filter/Spatial-js.html>. And send this filter through a WFS protocol. See the wfs-protocol.html example to know how to create a WFS protocol. To send the filter you'd use protocol.read({filter: yourSpatialFilter}).
