Maybe you should ask the spammers. :) Seriously, though, there are basically two options: VRFY (which many servers have disabled for just this reason); and starting a fake message and checking for recipient errors. Still, a lot of people don't reject mail for bum users, choosing instead to accept the mail and bounce it -- again, for precisely this reason.
