They explain it in a lot of detail in the Slashdot FAQ. Basically, your karma is a number in the database, and as you do positive things it goes up, and as you do negative stuff it goes down. Submitting stories, getting modded up on comments you post, and meta-moderating improve karma. Anonymous Cowards post at 0 points, normal users post at 1, and those with excellent karma post at 2 points. That's the gist of it, and I don't know any of the nitty gritty details.
