I wouldn't do more than use a lint free cloth, like a microfiber cloth and wipe the area of the speaker. I know some folks have mini vacuum cleaners they use on their braille displays to keep dust out of the braille cells, perhaps something like that...
Otherwise, if there is an Apple store anywhere near you, having it check out by a technician.
