I see a large difference, personally. I have a 768Kbps DSL line, but the path between my home and the office is often congested, and the TightVNC compression makes a noticable difference. The local cursor is also really helpful -- it doesn't affect the functioning of the connection that much, but psychologically it makes it feel more responsive.
