Can you boot off your installation DVD? That will tell you pretty quickly if its a hardware failure. If you can't then you probably need to talk to a Service Tech. If you can then you can use the options within the installation DVD to roll back to your last Time Machine backup and see if, once you have restored that, your machine will boot.
