I'm going to echo Mike's sentiment slightly differently: Building a successful site like Twitter, Facebook (or iTunes) takes great developers and good practices first and foremost. That's going to determine success more so than your choice of language (e.g: Java, Scala) or platform (e.g: WebObjects)...
