I'd say that depends on how often you (would) run defrag
Which should depend on how often the files on your partitions become so
fragmented that the system becomes not only noticeably, but also annoyingly
slower.
And the benefits you would expect to get from a defragment process as
opposed to any other process

My system 3 x 250Gb at the moment

My OS is on a separate drive from most software, so I rarely defrag the OS
drive
no more that twice in a year after I take, and verify backups of that
partition
( I do, more frequently, move the few 'user' files from, and then back to
the OS drive)

I have a separate partition for each users files including email and temp
internet stuff
These are subject to so much 'churn, that it isn't worth defragging.
( Occasionally move user stuff to a backup, delete all appropriate stuff,
and move the user 'data' files back, followed by the email stuff )

There is another partition for applications software
The applications rarely get changed so they rarely get fragmented
( not worth defrag effort)

There are other partitions for bulk file sets.
The bulk sets are occasionally moved to alternate partitions
Infrequent access to files means that fragmentation has little effect

So, with appropriately organised and structured partitions
other than to manipulate the MFT do you need to have Diskeeper,
And then, what manipulation facilities does it provide?
