Muscle memory and practice are definitely factors, and tedium, but that could also be up to the fact that they insist on doing straight runs with no breaks. For the most part, while they do seem to use cues and some memorization, for the most part they try to rely on what they call normalization, or controlled input. Things like the set distance you move by punching a wall, getting hit, etc. to reduce uncertainty as much as possible, but that varies from game to game. The video on the Blind Brotherhood I posted goes into a bit more detail on their processes, and they often publicly post their guides and strategies online for others. For example, the Portal playthrough has his strats notes in the description [here], which includes all the movements, cues, and steps he does to complete the blindfolded playthrough, a whole blueprint.
