With Plus turned off and unplugged any voltage that you find will be from battery. BTW, the schematic shows the battery voltage going to several chips, unlike (all?) other vintage macs which have it going only to the PRAM/clock chip. Soooo, it there's no diode, and you had reverse voltage applied, it's possible that more/other chips got damaged.
