I believe the conversation continued with Lady Astor saying, "Sir, if you were my husband, I would poison your tea." Whereupon Sir Winston replied,"Madam, if you were my wife, I would drink it!"

And he also said that "a preposition is a thing never to end a sentence with". My favourite though is the reported exchange with Lady Astor:

Her ladyship -- "Sir, you are drunk!"
