Chek whether the dirmngr process is already running. If that is the case kill it and run gpg-connect-agent --dirmngr --verbose It should tell you that it is trying to start dirmngr and you might see the reason for a problem. Watch out for firewall warnings; the GnuPG components use TCP connections to localhost to emaulate Unix localsockets. The Windows firewall and other software may want to block them.
