Not without third-party software. Exchange will always use your primary smtp address as the reply address if you're using Outlook and an RPC connection to Exchange.
I've heard of people setting up additional mailboxes with the additional address as the primary, giving the user Send As permission on that mailbox, and setting it to forward all the email to the user's primary mailbox. You can also send as a secondary address through POP. From: Lynden A. The problem there are a handful of users that has to switch between ABC.Com and DEF.com as there reply address and if someone sends an e-mail to them on there DEF.Com Domain it will automatically use that as there reply address or the other way as well.
