Dunno about the 4000, but with the 8000ED, an unmounted 645 slide that looks flat in the (non-glass) holder measures a focus range of about 0.1 units with Vuescan. A focus error of 0.05 units is no problem, but an error of 0.1 is beginning to be a problem. So I take a few readings across the frame and pick the middle of the range. Fortunately, taking a focus reading with VueScan is quite quick. David J.
