Actually, if Play Through AU is checked, and AU Setup is configiured
properly, simply hitting play should load the sounds...

I notice a change in the way 2006b behaves, one that is helpful to  
me, and I don't remember reading about it in the documentation.  It  
may be there, but I didn't notice, and I think it's worth mentioning.

When GPO or Garritan JABB is loaded through the Native Instruments  
option, my documents open without the sounds loaded, allowing files  
to be opened and closed quickly for edits, changes, and printing  
without having to wait two minutes for instruments to load, if you  
forgot to turn them off the last time you closed the document.  If  
"Play through Native Instruments" is selected in the MIDI menu, you  
now must invoke "Native Instruments AU setup" to get the sounds to  
load.  You don't have to go through the setup again, just select it,  
and it loads.  It's just a different way of doing business, but  
probably better for me in the overall picture.
